Restriction digests of the clone give the following sizes (kb): PstI/SstI--3.25, 0.54; PstI--3.8; SstI--3.7; XhoI--uncut; KpnI/BamHI--3.2, 0.5; RsaI--2.25, 0.72 (doublet); XhoI--1.5, 1.0, 0.8. The insert contains EcoRII (4), MboI (2), NsiI (2), HinfI (2), XhoII (1), RsaI (1), TaqI (1), AvaII (1), and HaeIII (2) sites. The insert consists of 483 bp of cDNA, 53 bp of dG/dC, and 17 bp of poly(A). The insert encodes the complete prepolypeptide of 114 amino acids and includes 11 bp 5' noncoding and about 128 bp 3' noncoding sequence. There is no AATAAA polyadenylation signal although there is a AGTAAA. Detects human genomic restriction fragments of the following sizes (kb): EcoRI--3.4 (and a weaker band of 3.0 kb that may be artifactual); BamHI--3.1, 0.5; PvuII--2.8, 0.8, 0.75. |
