Because of the temperature sensitive repressor, heat shock for CaCl2 transformations should be at 32C for 10 min, followed by 1 min on ice. Add LB and grow at room temperature for 1-2 hours.
Selection for ampR is on LB + amp (100 ug/ml) plates incubated at 30-32C.
An isogenic host without N gene function is TAP106, ATCC 47075.
